NIH Funds $12.6M AI Project to Redefine Alzheimer's Research and Treatment

A major new research project aims to transform Alzheimer's disease studies using artificial intelligence. The National Institutes of Health (NIH) has awarded $12.6 million to fund AI4AD2, a multi-institutional effort led by Dr. Paul M. Thompson at the USC Stevens Neuroimaging and Informatics Institute. The initiative will combine brain imaging, genomics, and cognitive data to uncover new insights into dementia and its subtypes. AI4AD2 brings together ten leading research institutions with a team of principal investigators and co-investigators. Their goal is to refine how Alzheimer's disease is classified at a molecular level, using AI to identify distinct subtypes and tailor treatments accordingly.

The project will develop 'genomic language models' to analyse vast genomic datasets and pinpoint genetic markers linked to Alzheimer's risk and progression. By leveraging PreSiBO, an AI-driven drug discovery platform, researchers will also explore genome-guided drug repurposing for more effective therapies. To ensure broad applicability, the team will validate their AI tools across diverse populations. The study includes over 57 cohorts, representing 12 ethnic or racial groups from regions such as Africa (Nigeria, Ghana, South Africa), South Asia (India, Pakistan, Bangladesh), East Asia (China, Japan, Korea), and Hispanic/Latino communities (Mexico, Puerto Rico, Colombia). European, Middle Eastern, and other ancestries are also included to address global health disparities. An open science framework will underpin the project, encouraging data sharing and collaboration. This approach aims to accelerate discoveries and make findings accessible to researchers worldwide.

The AI4AD2 project will advance precision medicine for Alzheimer's by identifying subtype-specific molecular targets and pathways. With its focus on inclusivity and open science, the initiative seeks to improve diagnostics and treatments for diverse populations. The NIH funding will support these efforts over the coming years.
